چکیده

Background: Human epidermal growth factor 2 (HER2) is known to be an important prognostic in breast cancer. Numerous studies have shown HER2+ cancers reduced overall survival and recovery time, as well the efficacy of anti-HER2 therapies along with chemotherapy improving disease outcomes. For this reason, it recommended that all patients cancer should evaluated for HER2 status. This study aimed assess gene amplification by CISH method evaluating status immunohistochemistry (IHC) 2+ (equivocal) results. Methods: retrospective cross-sectional examined based on Chromogenic situ hybridization (CISH) 280 carcinoma samples initial score IHC. The relationship between hormone receptors (estrogen progesterone) Ki67 level was also investigated. Results: In sixty (21.4%), amplified method. majority (215, 76.8%) were negative 5 (1.8%) indeterminate. No significant observed amplification, estrogen receptor (p=0.143), ki.67 protein (p=0.977). There a inverse progesterone positivity (p=0.007). Conclusion: These results demonstrate helpful equivocal positive (amplified) about 21.4% them.

Analysis of HER2 gene amplification using Differential PCR in breast cancer patients of Isfahan Province

Background: Amplification of HER2 is seen in 20-30% of breast cancer cases. Measurement of HER2 gene amplification appears to be of vital importance in planning the treatment schedule for patients with breast carcinoma. The aim of our study was to evaluate HER2 amplification status in malignant and benign breast tumors by differential PCR (dPCR). Materials and Methods: The genomic DNA was ex...

Evaluation of Immunohistochemistry-Equivocal (2+) HER2 Gene Status in Invasive Breast Cancer by Silver DNA in Situ Hybridization (SISH) and its Association with Clinicopathological Variables

Background and Objective:Determination of HER2 gene is crucial in breast carcinoma management and prognosis, as HER2 alterations are linked to a shorter disease-free period, overall survival and resistance to tamoxifen anti-estrogen therapy and other chemotherapy regimens, regardless of the nodal or hormone receptor status. This study aimed to...

Determination of HER2 Gene Amplification using Chromogenic in Situ Hybridization (CISH) in Iraqi Patients with Breast Carcinoma

Background: Breast cancer is the most common malignancy among women worldwide. The human epidermal growth factor receptor (HER) is a transmembrane receptor in the epidermal growth factor (EGF) receptor family located at the long arm of human chromosome 17 (17q12). Diagnostic assays for HER2 in breast cancer provide important prognostic information and independently help guide management by ident...

Activating HER2 mutations in HER2 gene amplification negative breast cancer.

UNLABELLED Data from 8 breast cancer genome-sequencing projects identified 25 patients with HER2 somatic mutations in cancers lacking HER2 gene amplification. To determine the phenotype of these mutations, we functionally characterized 13 HER2 mutations using in vitro kinase assays, protein structure analysis, cell culture, and xenograft experiments. Seven of these mutations are activating muta...
